After more than twenty years of use Good Laboratory Practice, or GLP, has attained a secure place in the world of testing chemicals and other “test items” with regard to their safety for humans and the environment. Gone are the days when the GLP regulations were hotly debated amongst scientists in academia and industry and were accused of stifling flexibility in, imaginative approaches to, and science-based conduct of, all kinds of studies concerned with toxic effects and other parameters important for the evaluation and assessment of products submitted for registration and permission to market. The GLP regulations have developed from rules on how to exactly document the planning, conduct and reporting of toxicity studies to a quality system for the management of a multitude of study types, from the simple determination of a physical/chemical parameter to the most complex field studies or ecotoxicology studies. At the same time the term “Good Laboratory Practice” has become somewhat ofa slogan with the aim to characterise any reliably conducted laboratory work.

Alike all other hill dwellers of the world, shifting cultivation, commonly known as 'Jhum', is the traditional agronomic practice among the indigenous people of hilly tracts of Bangladesh. This age old cultivation system affects, mainly negatively, on soil environment and worsens the upland watershed health. For investigating this hypothesis, a study was conducted at three pair sites viz: Simina Para, Kark Mohajon Para and At- mile area of Sadar upazila under Khagarachari district to determine the effects of shifting cultivation on soil environment. Both soil physico-chemical properties and the biochemical and biological properties of soil microbes were studied. The differences in microbial population count and population composition may not be universal for all shifting cultivation sites; however, it explains the unavoidability in the variations of the biological and biochemical characteristics of soil microflora in shifting cultivated areas and emphasizes the necessity of taking this factor into consideration in management of soils in fellow lands after shifting cultivation.

The border between Finland and Russia offers a unique opportunity for evaluating human impact on agricultural environments. Large areas of Karelia across the border were under similar management regimes while belonging to Autonomous Finland and Finland, but after the Second World War (1944), Karelia was divided by the present border. Since then, the history of agricultural management differed between adjacent areas in Finnish and Russian Karelia. In Finland changes in agricultural practices during the 20th century have led to a dramatic reduction in the number and area of semi-natural grasslands. The maintenance of these species rich plant communities can be ensured by grazing or hay cutting, but half of the remaining sites in the province of South Karelia were abandoned. In addition, only a fraction of the sites, still in use, were sustainably managed. In Russian Karelia changes in agricultural practices have taken place on a much smaller scale. This book summarizes the effects of different agricultural practices on the abundance of semi-natural grasslands and their vegetation in the border district of Finland and Russia.

Regarding a strategy of expressing foreign genes in plants, two diverged RMV-based expression vectors containing a green fluorescent protein were cloned into the binary vector pPZP200 for subsequent agroinfiltration work. An infectious full-length clone of RMV and three RMV-based mutants already existed. They vary depending on which of the two putative ORFs is inactivated. An additional mutation site of one of these mutation constructs was discovered and reverse mutated. After the inoculation of the constructs on N. benthamiana and N. tabacum, the analysis revealed distinct effects in virus accumulation and symptom characteristics of the three mutation constructs. These results differ from the ones of the wild type clone. Accordingly, all three mutants show both lower virus accumulation levels and lesser symptom characteristics. Further, the results from N. benthamiana indicate an ability to reverse mutate the -4K virus. In addition to the description and the analysis of these results, the potential role of the two putative ORFs will be discussed and an outlook on further investigations will be given.
